摘  要:目的检测接受体外受精-胚胎移植(IVF-ET)的不孕患者取卵日卵泡液中白血病抑制因子(LIF)水平,探讨其与卵母细胞成熟度及甾体激素水平关系。方法采用ELISA法检测61例IVF-ET惠者取卵日血清及卵泡液中的LIF水平、采用放射免疫法(RIA)检测卵泡液中雌二醇(E2)、孕酮(P)、睾酮(T)水平,并记录每份卵泡液中的卵母细胞成熟度。结果(1)卵泡液中的LIF水平为(403.45±128.15)pg/ml,显著高于血清中LIF水平[(3.52±1.54)pg/ml)](P<0.001)。(2)成熟卵细胞卵泡液中LIF水平为(529.5±218.17)pg/ml,显著高于不成熟组[(194.70±92.65)pg/ml] (P<0.001)及过熟组[(321.42±131.35)pg/ml](P<0.05)。(3)卵泡液中LIF水平与P成正相关(γ=0.343,P=0.007),与E2、T无相关性。结论卵泡液中高水平的LIF可能为卵巢源性,其与卵母细胞的成熟度有密切关系,并且与卵泡液中的甾体激素有一定的相关性。Objective To detect the concentration of leukaemia inhibitory factor(LIF) in human follicular fluid(FF) in vitro fertilization and embryo transfer(IVF-ET) ,and to evaluate its relationship to oocyte maturity and sex hormones levels. Methods LIF concentrations in FF and serum samples obtaining from 61 patients undergoing IVF-ET were measured with ELISA. The levels of estradiol(E2 ),progesterone (P),testosterone (T) were determined by radioimmunoassay. The maturity of each oocyte was identified. Results The LIF mean concentration in FF[(403.45±128. 15) pg/ml] was significantly higher than that in serum[(3.52±1.54) pg/ml] (P〈0. 001). The LIF mean concentration in FF with mature oocytes [(529.56±218. 17) pg/ml] was significantly higher than those with premature oocytes [(194.70±92. 65) pg/ml] (P〈0. 001) and overmature oocytes [(321. 42±131. 35) pg/ml] (P〈0. 05). In FF, the concentration of LIF was positively correlated to P level (γ=0. 343, P〈0. 05). No positive correlation was found between the level of LIF and the level of E2 and T in FF.Conclusion Higher concentration of LIF exist in FF and it may have a physiological role in promoting oocyte development and maturity.
