摘　　要：目的:通过比较重度子痫前期和正常孕妇胎盘mRNA表达水平的差异,发现子痫前期易感基因。方法:应用荧光mRNA差异显示(FDD)技术,包括2种锚定引物和4种随机引物,共8种组合,测定重度子痫前期患者和正常孕妇胎盘组织基因表达水平,对获得的差异基因片段进行PCR扩增、克隆及测序,测序结果提交GenBank熏经BLAST软件检索并进行同源性分析。结果:重度子痫前期患者胎盘高表达基因片段19条,序列分析表明其中两条分别与丝-苏氨酸蛋白磷酸酶2A(PP2A)β催化亚基及ATP结合盒转运子G2(ABCG2)基因同源性达99%,还有2条cDNA序列与GenBank中已知序列无高度同源性,申请登录GenBank,登录号分别为CV998051、CV998052。结论:重度子痫前期患者胎盘中PP2A、ABCG2高表达,可能与本病的发生、发展有关。Objective： To compare the difference of mRNA expression in placenta between severe preeclampsia and normal pregnancy, and find the susceptible factors of preeclampsia.Methods： The levels of gene expression in placenta of severe preeclampsia and normal pregnancy were compared by fluorescent differential display reverse transcription polymerase chain reaction （FDD-RTPCR） including two sets of anchor primers and four sets of arbitrary primers,totally eight primer combinations.Gene fragments obtained were amplified by PCR, then were cloned and sequenced.The sequence results were compared with GenBank database by BLAST software for homologous analysis. Results： Nineteen highly expressed gene fragments in placenta of severe preeclampsia were found. Two of them were respectively highly homologous to human protein phosphatase 2A （PP2A）catalytic subunit-beta and ATP Binding Cassette transporter G2 （ABCG2） gene,and the homologies were 99%. Two cDNA frag ments which have low homology with known gene were discovered,and the sequence data were submitted to GenBank with accession number CV 998051 and CV 998052. Conclusion： High expressions of PP2A and ABCG2 in placenta of severe preeclampsia were found by FDD, and they may be related to the occurrence and development of this disease.
