高效液相色谱法测定盐酸美金刚烷胺片的含量

摘　　要：目的建立HPLC测定盐酸美金刚烷胺片含量的方法。方法采用HypersilODS柱(250mm×4.6mm,5μm)为固定相,流动相为0.33%磷酸氢二钾溶液乙腈(40∶60,v/v);检测波长为197nm;流速为1.5mL·min-1;柱温30℃。结果本方法在0.5～2.0mg·mL-1呈良好线性关系(r=0.9999),进样重现性RSD为1.08%(n=5),平均回收率为98.50%,日内RSD为1.68%(n=5),日间RSD为1.82%(n=5)。结论本方法简便、快速,结果准确可靠,可用于盐酸美金刚烷胺片的含量测定。OBJECTIVE To establish an external standard HPLC method for the determination of memantine hydrochloride. METHODS Hypersil ODS column was used and 0.33% potassium Dihydrogen Phosphate-Acetonitrile （40：60, v/v） was used the as mobile phase, and detected at 197 nm The flow rate was 1.5 mL·min^-1 , and the column temperature was 30 C. RESULTS The linear range of the method was 0. 5-2.0 mg·mL^-1 （r=0. 9999）, RSD=1.08% （n=5）, and the average recovery was 98. 50%. The precision RSD within a day and between dayswere 1.68% （n=5） and 1.82% （n=5）. CONCLUSIONS The method is simple, fast and accurate for the analysis of memantine hydrochloride.
