摘 要:目的:探讨离体SD大鼠主动脉对一定浓度胰岛素的反应性改变及其主要机制。方法:采用血管环灌流装置,对比观察离体SD大鼠胸主动脉环在内皮存在条件下,由去甲肾上腺素(NE)引发收缩后, 未加干预或施加干预(L-NAME或5-HD抑制剂处理)对Ins反应的变化。结果:Ins(120 U/L)对NE引发收缩后的离体SD大鼠胸主动脉环具有舒张作用,血管舒张率为:(24±8)%,P<0.01,用L-NAME或5-HD抑制剂处理后,Ins对离体SD大鼠胸主动脉环的舒张作用明显减弱,血管舒张率分别为:(9±3)% 和(14±5)%,P<0.01。结论:Ins对离体SD大鼠胸主动脉环具有舒张作用,L-NAME或5-HD可阻断该舒张反应,故考虑Ins介导的血管舒张反应主要是内皮或NO依赖的舒张反应,少部分舒张反应是通过诱导mitoKATP通道开放来实现的。Objective: To investigate the vascular reactivity to the insulin on isolated aorta. Methods: The thoracic aortic rings with intact endothelium of male Sprague-Dawley rat were mounted on a bath system. Aortic rings were subjected to contractile concentration responses to NE followed by rclaxation to insulin(120 U/L),with and without preincubation for 15 minute with L-NAME or 5-Hydroxydecanoate(5-HD). vascular reactivety on aortic rings was measured using an isometric force transducer. Results: Insulin induced significantly vasorelaxation (percent of relaxation (24±8)%,P〈0.01). After preincubation with L-NAME or 5-Hydroxydecanoate, both of the relaxation responses to insulin were impaired. (L-NAME (9±3)% P〈0.01 vs 5-HD (14± 5)%, P〈 0.01) Conclusion: Thc relaxatation of insulin partly is endothelium or NO-dependent, The responses was reduccd with the inhibition of mito K (ATP) channel, suggesting that the vasorelaxant mechanisms are in part dependent to mito K (ATP) channel opening.
