摘 要:目的:探讨脂联素对体外培养大鼠肝脏细胞葡萄糖激酶与磷酸烯醇式丙酮酸羧激酶活性影响,及其在胰岛素抵抗中的意义。方法:以BRL肝脏细胞株为细胞模型,分别用10μg/ml、30μg/ml、50μg/ml脂联素处理细胞,用6-磷酸葡萄糖脱氢酶偶联比色法、乳酸脱氢酶偶联比色法检测大鼠肝脏细胞葡萄糖激酶与磷酸烯醇式丙酮酸羧激酶活性。结果:与对照组比较,磷酸烯醇式丙酮酸羧激酶在脂联素处理24h后活性明显降低,且随脂联素浓度升高作用越明显,P<0.05。葡萄糖激酶在脂联素处理24小时后,活性与对照组比较没有显著性差异,P>0.5。结论:脂联素对葡萄糖激酶无明显的调节作用,而对于磷酸烯醇式丙酮酸羧激酶具有显著的抑制作用。脂联素通过对肝细胞磷酸烯醇式丙酮酸羧激酶活性的调节,降低肝脏葡萄糖异生,减少肝糖输出,改善胰岛素抵抗。Objective :To investigate the effect of adiponectin on enzymatic activity of glucokinase and phosphoenolpyruvate carboxykinase in cultured rat's liver cells. Methods : The BRL was used as cell model. The rat liver cells were incubated with adiponeetin of three different concentrations ( 10μg/ml, 30μg/ml and 50μg/ml). Controlled cells were incubated without adiponectin. Enzymatic activity of GK was investigated by using a standard G6P - coupled assay. Enzymatieal activity of PEPCK was assessed by using a standard LDH- coupled assay. Results :Compared with the control group, treated group with adiponectin of all concentrations for 24h had no significant effect on enzymatic activity of glueokinase. Enzymatic activity of phosphoenolpyruvate earboxykinase was significantly decreased even in the lowest concentration of adiponectin,and along with increased in adiponectin concentration enzymatic activity of phosphoenolpyruvate earboxykinase decreased more significantly. Oonclusion :Adiponectin has no significant effect on ezymatic activity of glucokinase. However adiponcctin decreases enzymatic activity of phosphoenolpyruvate carboxykinase significantly. Through this effeet, adiponectin deereaes liver glucose production and improves insulin resistance.
