摘  要:用异硫氰酸荧光素衍生普萘洛尔,衍生产物用毛细管电泳分离,激光诱导荧光方法检测.在含有10 mmol/L β-CD,50 mmol/L 硼砂(pH=9.0)的溶液中,普萘洛尔对映体得到了基线分离.本方法线性范围宽(R型:2.0×10-3~1 mg/L), 灵敏度高(检出限均为1.24 μg/L),可用于活体中普萘洛尔对映体的药代动力学研究.Propranolol (PPL) can react with fluorescence-5-isothiocyanate (FITC) in an appropriate condition, then derivative FITC-PPL is achieved. FITC-PPL was baseline separated using 10 mmol/L β-cyclodextrin as chiral selector and 50 mmol/L borate solution as buffer. The uncoated fused-silica capillary of 75μm i. d.and 57 cm (effective length 50 cm) was used. The capillary temperature was maintained at 20℃. Samples were inejcted with pressure at 3.45 kPa for 3 s and separated at 18 kV. Both R-PPL and S-PPL had low detection limits ( 1.24μg/L). The linear range of R-PPL was 2.0 × 10^-3 - 1.0 mg/L. The method offers a high sensitivity and broad linear range, and it can be used to do the research of the pharmacokinetics of PPL enantiomers.
