摘  要:目的制备免疫载药毫微粒HILE6-PLA(5-Fu)-NP,体外评价其表征、免疫学特性及对晶状体上皮细胞(LECs)增殖的抑制作用。方法将抗人LECs膜单克隆抗体HILE6与5-Fu聚乳酸毫微粒采用碳二亚胺法相连接制备免疫毫微粒。扫描电镜观察微粒形态;间接免疫荧光法评价其免疫特异性;建立LECs、角膜基质细胞培养模型,体外检测不同质量浓度免疫微粒和单纯载药微粒对细胞作用2h后的杀伤作用。结果免疫毫微粒保持对LECs的免疫特异性,对LECs具有很强的杀伤作用,明显高于同质量浓度毫微粒,62·5μg/ml作用2h抑制率可达到92·3%,撤药后观察时间延长到7d时的抑制作用高于或不低于3d时的效果。使用药物质量浓度<12·5μg/ml对角膜细胞相对较安全。结论该免疫毫微粒可有选择性的,长时间杀伤LECs,预防后囊混浊。Objective To prepare an immunonanoparticles HILE6-PLA (5-Fu)-NP and evaluate the immunological characters and the toxicity to lens epithelial cells (LECs) in vitro. Methods The immunonanoparticles were made by crosslinking LEC monoclonal antibody HILE6 to PLA nanopartieles loading 5-fluorouracil PLA (5-Fu)-NP. The morphology of HILE6-PLA(5-Fu)-NP was observed by scanning electrical microscope. The immunological characters of HILE6-PLA (5-Fu)-NP was analyzed by indirect immunofluorescenee. LECs and cornea stroma cell culture system were used for the testing of the cytotoxity of HILE6-PLA(5-Fu)-NP and PLA(5-Fu)-NP with different dosage in vitro and divided into 3 groups: LECs with PLA (5-Fu)-NP( Group A ) ;LECs with HILE6-PLA(5-Fu)-NP( Group B ) ;cornea stroma cell with HILE6-PLA (5-Fu)-NP( Group C ). Results HILE6-PLA(5-Fu)-NP remained the immunological activity and had stronger potential to inhibit LECs proliferation than PLA (5-Fu)-NP at the same concentration, A 92, 3% inhibition was observed at 2 hours after exposure of LECs to 62. 5μg/ml of immunonanoparticles. The inhibitory effect at the 7th day was equal to or better than that at the 3rd day after withdrawal of the HILE6-PLA(5-Fu)-NP, 12.5μg/ml or below HILE6-PLA (5-Fu)-NP had no obvious effect on the cornea stroma cells,Conclusion The HILE6-PLA (5-Fu)-NP can prevent posterior capsular opacification through selectively and long-time killingLECs.
