摘 要:目的探讨虎参痛风胶囊(Hus)对高尿酸血症小鼠T淋巴细胞亚群的影响。方法小鼠按随机化分组原则分为空白组、别嘌呤醇组、模型组及5%,2.5%,1.25%虎参痛风胶囊组,灌胃给药6d后,采用腹腔注射黄嘌呤一次造模,30min后摘眼球取血,尿酸酶比色法测定血尿酸;处死小鼠,取脾脏,剪碎后,制备成单细胞悬液,单色法应用流式细胞仪测定CD3+,CD4+,CD8+百分比及CD4+/CD8+比值。结果模型组血尿酸明显升高(与空白组比较,P<0.05),而别嘌呤醇组和5%,2.5%,1.25%虎参痛风胶囊组能明显降低血尿酸(与模型组比较,P<0.05)。应用流式细胞仪检测脾脏T淋巴细胞分型结果显示模型组、别嘌呤醇组和空白组CD3+,CD4+,CD8+及CD4+/CD8+均无显著性差异;且5%虎参痛风胶囊组与模型组、空白组、别嘌呤醇组相比较亦无显著性差异。结论Hus具有明显的降低高尿酸血症小鼠血尿酸的作用,优于别嘌呤醇。急性高尿酸血症和Hus对脾脏T淋巴细胞亚群无明显影响。Objective:To elucidate the influence in the subpopulations of T lymphocyte administrated Hus in mice. Methods:Adopt 10% xanthine i. p. in mice to duplicate hyperuricemia model,and administrate drugs for 6 days beforehand.Then, to detect serum urate concentration by uricase chromatometry, and to measure the percentage of CD3^+,CD4^+,CD8^+ and CD4^+/CD8^+ ratio of spleen T lymphocytes by monochromic flow cytometry (FCM). Results:5% ,2.5% ,1.25% Hus group and allopurinol group had significant differentiation respectively compared with model group (P〈0.01,n=8),and were lower than the model group, respectively. And 5%,2.5% Hus group had significant differentiation respectively compared with allopurinol group. It denoted that different dosage Hus can respectively reduce serum urine content in hyperurieemia of mice as similar as allopurinol, and even be lower than normal level. In contrast, there were no significantdifferences between allopurinol group, model group and normal group in the percentage of CD3^+、CD4^+、CD8^+ cells and CD4^+/CD8^+ ratio in spleen lymphocytes,nor were there significant differences between normal group ,model group, allopurinol group and 5% Hus group in the percentage of CD3^+,CD4^+,CD8^+ cells or CD1^+/CD8^+ ratio of spleen lymphocytes. Conclusion: Hus can obviously significant influence in spleen T lymphocyte subpopulations.
