摘 要:探讨止喘胶囊对哮喘大鼠气道TGF-β1信号转导的干预作用。通过长期反复给予致敏大鼠吸入不同浓度的卵蛋白,建立大鼠慢性哮喘模型。40只大鼠随机分为5组:正常对照组、模型组、中药组、西药组和中西结合组。原位杂交技术检测Smad-2mRNA、Smad-7mRNA表达水平。结果:各治疗组均可下调Smad-2mRNA的表达,与模型组比较有显著性差异(P<0.05),其中尤以中西医结合组疗效最佳,中药组与西药组效果相当(P>0.05)。中药组可显著上调Smad-7mRNA表达水平,与模型组比较差异显著(P<0.05);而西药组与模型组比较,无显著性差异(P>0.05)。结论:止喘胶囊能通过下调Smad-2mRNA。上调Smad-7mRNA的表达,阻断TCF-β1的信号转导,从而阻断哮喘气道重塑的发展。To explore the mechanism of sigual conduction of TGF-β1 on the airway remodeling of asthmatic model rats treating with ZhiChuan capsule, the model was established by sensitized and activatived with different density ovalbunin (OA) through prolonged and repeated exposure for 8 weeks. The rats were randomly divided into normal control, model group, Chinese medicine group, western group, and Chinese-westero group. General histological changes were observed by hematoxylin and eosin stained sections. The express of Smad-2 mRNA, Smad-TmRNA in the airway were examined by ISH. Statistical comparison was performed by ANOVA followed by Fisher LSD test. Results:( 1 ) All of the treating groups could inhibit Smad-2 mRNA expression, especially in the Chinese-westem group; (2) Chinese medicine group could up regulate Smad-2 mRNA expression, and the western group had no such effect. Conclusions ZhiChuan capsule could up regulate Smad-7mRNA expression, down regulate Smad-7mRNA expression, and inhibit signal conduction of TGF-β1, then suppress airway remodeling of bronchial asthma.
