摘  要:目的建立测定人血清中卡马西平及其代谢物10,11-环氧化卡马西平浓度的方法.方法采用高效液相色谱法,血样用乙醚提取,以艾司唑仑为内标,色谱柱为Kromasil C18柱(150 mm ×4.6 mm,5μm),流动相为乙腈-水(1:2),检测波长215 nm,流速1.2ml·min-1.结果血清中卡马西平、环氧化卡马西平线性范围分别为1.0~20.0 mg·L-1(r=0.999 9)、0.1~5.0 mg·L-1(r=0.999 4),平均回收率接近100%,日内、日间RSD均小于5%.结论本法快速、简便、准确,适用于临床常规监测需要.Aim To determine carbamazepine, carbamazepine-10,11-epoxide in serum by high performance liquid chromatography (HPLC). Methods Serum sample was extracted with 5ml aether, Estazolam was selected as an internal standard, An UV detector (215 nm)and Kromasil C18 column (150mm×4.6mm,5μm)were used. The mobile phase consisted of acetonitrile-water( 1:2 ). The flow rate was 1.2mg·L^-1. Results The linear range of carbamazepine and carbamazepine-10,11-epoxide were 1.0-20.0mg·L^-1(r=0.999 9) ,0.1-5.0mg·L^-1( r = 0.999 4 ) ,. The mean recovery were nearly 100% ,. The relative standard deviation (RSD) of inter-day and intra -day for carbamazepine and carbamazepine-10,11-epoxide were less than 5%. Conclusion The method is accurate, sensitive and more suitable for clinical routine monitoring.
