摘 要:目的探讨RGDPe(Ac-Arg-Gly-Asp-NHCH2CH2C6H5)和RGDmpe(Nα-acetylarg-gly-asp-p-methoxylphenylethylamine)肽类血小板聚集抑制剂抑制动物体内动脉血栓形成和溶栓的生物活性。方法制备Wistar大鼠血栓模型,以血栓干湿重和光镜、扫描电镜等指标观察RGDPe和BGDmpe预防血栓和溶栓作用。结果RGDPe、BGDmpe抑制动脉血栓形成作用相似(P>0.05),明显强于生理盐水对照(P<0.001);RGDPe、RGDmPe预防血栓形成作用优于BGDS(P<0.01);溶栓作用均明显低于本实验用量尿激酶(P<0.05);当它们分别合用尿激酶时,其溶栓作用与单用二者之一明显增强(P<0.01)。结论RGDPe、RGDmPe都能有较强的抑制动脉血栓形成和一定的溶栓作用,与尿激酶有较好的协同溶栓作用。Objective To explore the effect and biological activities of RGDPe( Ac-Arg-GIy-Asp-NHCH2CH2C6H5 ) and RGDmPe ( N(-acetyl Arg-Gly-Asp-p-methoxyl phenylethylamine) on the prevention of thrombosis formation and thrombolysis. Methods Wistar rat model of thrombus was used to observe the preventive roles of RGDPe and RGDmPe in thrombosis formation and thrombolysis with the weight of both dried and wet thrombus, the optical microscope and scanning electron microscope. Results RGDPe and RGDmPe have the similar activity to inhibit thrombus formation(P 〉 0.05), but stronger than saline group( P 〈 0. 001 ). RGDPe and RGDmPe are significantly more effective than RGDs in inhibiting thrombosis( P 〈 0.01 ), but less effective than urokinase( 12 500 U/kg) (P 〈 0.05 ) in thrombolysis. They are far more effective when each of them combined with urokinase than themselves respectively in thrombolysis (P 〈 0.01 ). Conclusions Both RGDPe and RGDmPe could have biological activities of inhibiting thrombosis and promoting thrombolysis, and play the synergetic role combined with urokinase.
