摘 要:目的:许旺细胞能够促进损伤的周围神经的再生过程。应用许旺细胞与组织工程材料联合培养,构建人工神经修复周围神经损伤,具有广阔的应用前景。因此,探讨许旺细胞的培养方法以及许旺细胞与三维支架的联合培养对组织工程化人工神经的构建具有重要意义。资料来源:应用计算机检索Medline1984-01/2004-01中有关许旺细胞培养以及组织工程材料的文章,检索词为“Schwanncellsculture,tissuengineeringmaterials,associationculture”,并限定文章语言种类为“English”;同时应用计算机检索中文期刊全文数据库和万方数据库199601/2004-12中有关许旺细胞培养以及组织工程材料的文章,检索词为“许旺细胞培养,组织工程材料,联合培养”,限定文章语言种类为中文。资料选择:对资料进行初审,重点选择介绍许旺细胞培养方法以及许旺细胞与三维支架联合培养的文献,对非随机试验予以排除。资料提炼:在所有检索到的文献中,排除非随机试验的文献以后,最终选择24篇最为相关的作为参考文献。其中有1篇重点介绍许旺细胞的作用,5篇重点介绍许旺细胞的培养方法,其余的主要研究许旺细胞与三维支架的联合培养。资料综合:许旺细胞的培养方法包括组织块法和酶消化培养法。可与许旺细胞进行联合培养的支架材料包括聚酯纤维、聚羟基乙酸、聚乳酸、聚羟基乙酸和聚乳酸共聚物、ZQ膜、壳聚糖等等。这些支架材料为人工神经的构建提供了丰富的原料,为应用组织工程化人工神经修复受损的周围神经奠定了基础。结论:应用组织块法和酶消化培养许旺细胞的技术已经基本成熟。对于那些可与许旺细胞进行联合培养的支架材料的研究,应当尽快由现今单纯的体外培养逐渐向动物实验过渡,为早日研制出合格的人工神经做进一步的努力。OBJECTIVE:Schwann cells can protnole the process that injured peripheral nerves regenerate.Using Schwann cells for association culture with tissue engineering materials and fabricating artificial nerve to repair injured peripheral nerves have extensive applied prospect.Therefore,the study of the culture method of Schwann cells and the association culture between Schwann cells and three-dimensional seaffolds is very important to fabricate tissue engineering based artificial nerve.DATA SOURCES:Using the key words of“Schwann cells culture.tissue engineering materials,association culture”,the artieles about the culture of Schwann cells and tissue engineering materials between January 1984 and January 2004 were searched for in Medline by using computer.The language of the articles was limited to English.Meanwhile,using the key words of“Schwann cells culture,tissue engineering materials,association culture”,searched for the related articles between January 1996 and December 2004 in China Journal Full-text Database(CJFD)and Wanfang database with the language limited to Chinese.STUDY SELECTION: After the first-time examination, the articles about the culture methods of Schwann cells and association culture between Schwann cells and three-dimensional scaffolds were selected mainly, while the nonrandomized experiments were excluded.DATA EXTRACTION: Among all the articles, 24 literatures were collected as relative reference literatures after excluding the articles which were nonrandomized experiments. Among all the references, 1 article was mainly about the effects of Schwann cells, 5 articles were about the culture methods of Schwann cells, and the others were relevant to the association culture between Schwann cells and the three-dimensional scaffolds. DATA SYNTHESIS: The Schwann cells could be cultured by tissue block method and enzymes digesting method.
