摘  要:目的观察肌凝蛋白诱导Balb/c小鼠发生自身免疫性心肌炎后小鼠免疫功能的变化。方法以猪心肌中提取并纯化的肌凝蛋白免疫Balb/c小鼠,建立自身免疫性心肌炎模型。流式细胞仪检测小鼠外周血和脾淋巴细胞CD3+CD4+、CD3+CD8+、Th1、Th2及TCRVβ8.1&8.2和TCRVβ10的变化,酶联吸附试验观察外周血TGF-β1和抗肌凝蛋白抗体的变化3。H-TdR检测小鼠脾脏单个核细胞对肌凝蛋白刺激的增殖反应。结果94%(15/16)小鼠在免疫后第18天出现不同程度的心肌炎病理改变,主要表现为心肌间质单个核细胞浸润;免疫鼠血浆肌钙蛋白I水平升高,TGF-β1水平显著下降,并出现较高滴度的抗肌凝蛋白自身抗体。免疫小鼠外周血CD3+CD4+细胞较正常对照小鼠显著增高。外周血和脾脏Th1、Th1/Th2及TCRVβ8.1&8.2阳性T淋巴细胞显著增加;小鼠脾脏单个核细胞对肌凝蛋白的增殖反应较对照组明显增加。结论猪心肌肌凝蛋白可以诱导Balb/c小鼠出现典型的自身免疫性心肌炎,是一种以Th1细胞介导为主的自身免疫反应。Objective To evaluate immune changes of experimental autoimmune myocarditis in Balb/c mice induced by immunization of myosin. Methods Cardiac myosin was isolated and purified from porcine heart. Experimental autoimmune myocarditis was induced in Balb/c mice immunized with cardiac myosin. CD3^+ CD4^+ T lymphocytes, CD3^+CD8^+ T lymphocytes, Th1 , Th2, TCRVβ8. 1&8.2+ T lymphocytes, TCRVβ10+ T lymphocytes in mice peripheral blood and spleen were analyzed by flow cytometry. Plasma TGF-β1 level and myosin antibody level were determined using ELISA. ^3H-TDR incorporation test was done to determine the proliferative response of mice spleen mononuclear cells stimulated with myosin. Results On 18d after the first immunization with cardiac myosin, 94%(15/16) of the Balb/c mice developed myocarditis with pathological feature of infiltration of mononuclear cells. High titer of myosin-specific autoantibody and elevated troponin I level were detected in plasma of immunized mice. CD3^+ CD4^+ T lymphocytes in peripheral blood of immunized mice increased significantly compared with the control. Th1, Th1/Th2 and TCRVβ8.1+8.2+ T lymphocytes in peripheral blood and spleen of immunized mice increased significantly compared with the control. Plasma TGF-β1 decreased significantly in immunized mice. The proliferative response of immunized mice increased significantly compared with the control group. Conclusion Cardiac myosin isolated from porcine is capable of inducing autoimmune myocarditis in Balb/c mice. The results suggest that experimental autoimmune myocarditis is mainly caused by Th1-mediated response.
