摘 要:目的探讨中国地鼠糖尿病发作时外周血浆儿茶酚胺及血管紧张素II与慢性应激的关系及作用机制。方法中国地鼠161只,采用三因素二水平析因设计,共分8组,考虑雌雄性别均衡匹配;实验性应激选用震动,旋转和合并限制等方法,实验时间为1年。监测血糖,1年后,检测血浆肾上腺素(E),去甲肾上腺素(NE),胰岛素(INS),心肌血管紧张素II(AII)含量。并计算胰岛素敏感指数(ISI)。结果(1)慢性实验性应激和高情绪唤醒组血糖水平[(6.54±0.72)mmol/L]显著高于非应激组[(5.69±0.09)mmol/L](P<0.01),血浆胰岛素浓度和胰岛素敏感指数(4.48±0.69)显著低于非应激组(10.67±1.66)(P<0.01),但血浆肾上腺素,去甲肾上腺素,心肌组织血管紧张素II组间差异无显著性(P>0.05)。(2)情绪唤醒水平和笼架水平对血浆ISI,E,NE及心肌AII含量作用不显著(P>0.05)。结论慢性实验性应激和高情绪唤醒可以使中国地鼠血糖增高,但外周血浆E,NE,心肌组织AII,并不具有显著的作用,可能是慢性实验性应激的适应性表现。笼架水平对中国地鼠的糖代谢改变无显著影响。Objective To investigate the effect of chronic experiment stress and relate emotionahty arousal and cage level to plasma glucose level in Chinese hamster and explore the glucose metabolic mechanism of stress in the onset diabetes mellitus of Chinese hamster. Methods According to the design of a three-way ANOVA analyses, 161 three month-old Chinese hamsters were selected to assign into 8 groups and matched with sex. Experimental stressors were shaking, rotation and restraint lasting one year. After 1 year all rats were killed, and collected blood and heart samples and then detected insulin (INS) , epinphine (E) , noepinphine (NE) in plasma and heart angiotensin Ⅱ (AⅡ), and to calculate the insulin sensitivity by insulin sensitivity index [1/( BG INS)](ISI). Results ( 1 ) The glucose level was significantly increased in experimental stress animals ( P = 0. 001 ) ;the insulin concentrations in plasma and insulin sensitivity index was significantly lower( P〈 0.01 ) in experimental stress rats than non stress rat, but the NE, E, in plasma and All in heart were not significantly influenced by the experimental stress. (2) The effect of emotionality arousal and cage shelf in plasma INS, NE, E, and All in heart were no significance. Conclusion The glucose level of Chinese hamster was significantly increased in experimental stress and emotionality, but NE,E in plasma and All in heart were not significantly effect . The cage level is not significantly effect to glucose metabolic in Chinese hamster.
