摘 要:方法:采用三种方法对苹果褪绿叶斑病毒RT—PCR的特异DNA产物进行回收纯化。目的:针对不同情况,选择适宜的回收纯化方法。结果:用普通琼脂糖替代低融点琼脂糖,回收纯化后产物的浓度及纯度与低融点琼脂糖法基本一致,完全可以用普通琼脂糖替代低融点琼脂糖进行DNA片段的回收纯化,从而降低成本,简化操作。玻璃奶法的回收纯度明显高于低融点琼脂糖法和普通琼脂糖法,且更快速安全。是采用普通琼脂糖法还是采用玻璃奶法回收纯化DAN片段应以实际需要而定。Method:Three kinds of methods were chosen to extract and purify DNA fragments from agarose gel.Objective:To select the suitable methods to extract and purify DNA fragments.Results:the purity and concentration of DNA fragments extracted and purified by common agarose method was almost the same as that by low-melting agarose method,and the low melting agarose method could be substitued by the common agarose method.The purity and coneentration of DNA fragments extracted and purified by glass milk method was obwiously higher than that by the other methods.Meanwhile the glass milk method had the characteristics of safety and simplification.but the cost of it was five to six times higher than that of the common agarose method.
