摘  要:目的:探索新型肿瘤坏死因子(TNF)相关的凋亡诱导配体(TRAIL)对乙型肝炎病毒(HBV)感染后的免疫监视效应。方法:应用酶联免疫吸咐(ELISA)和噻唑蓝(MTT)比色法及流式细胞术检测52例HBV感染者和24例健康对照者血清中可溶性TRAIL(sTRAIL)的表达,在转染HBV全基因组并稳定表达病毒抗原的HepG2.2.15肝癌细胞系研究TRAIL在γ鄄干扰素(IFN鄄γ)的作用下对HepG2.2.15细胞的生长抑制效应和凋亡诱导效应。结果:HBV感染者的外周血sTRAIL的表达量明显高于正常对照组(P<0.001)。各组HBV感染者sTRAIL的表达量与丙氨酸氨基转换酶(ALT)的值呈正相关。而细胞学的研究结果显示,在IFN鄄γ的协同作用下,TRAIL能够显著抑制HepG2.2.15细胞的恶性增殖,并能够显著诱导HBV相关肿瘤细胞发生凋亡。结论:TRAIL是参与到HBV感染后所致疾病中的一个重要的免疫监视分子,TRAIL的作用效应极有可能直接影响疾病的预后与转归。Objective:To explore the immune surveillance effect of TRAIL, a new tumor necrosis factor-related apoptosis inducing ligand on the HBV related diseases. Methods: The expression of sTRAIL in 52 HBV infected patients and 24 healthy controls, the growth inhibitory effect and apoptosis inducing effect of TRAIL and IFN-У, in HepG2.2.15 hepatocarcinomatous cell line transfecting HBV were studied by ELISA, MTT and FCM assay, respectively. Results: The expression of sTRAIL in the HBV infected group was significantly higher than that in the healthy control group (P < 0.001). In the HBV infected group, the expression level of sTRAIL was positively related to the ALT value. Cellular study showed that with the cooperation of IFN-У, TRAIL could significantly inhibit the growth and induce apoptosis of the HBV related HepG2.2.15 cells. Conclusion: TRAIL is an important immune surveillance molecule that probably plays a key role in the prognosis of the HBV related disease.
