停服棉酚后恢复的大鼠精母细胞DNA合成的研究

摘  要:用细胞淘析分离技术分离出经棉酚处理后的大鼠粗线期精母细胞,用^3H-胸腺嘧啶核苷参入法检测棉酚对DNA合成的影响。结果表明,饲服棉酚3、4、6、7和8周后,大鼠粗线期精母细胞的DNA合成明显低于对照组,分别降低14%、26%、40%、42%和40%。本文对棉酚的作用机理进行了讨论。In this paper centrifugal elutriation technique was used to separate the repopulating pachytene spermatocytes from the rats recieved gossypol treatment and the normal pachytene spermatocytes from the normal rats,By means of in vitro measurement of the ration of 3H-TdR incorporation the activity of DNA synthesis in the repopulating pachytene spermatocytes was detected.The results show that following gossypol treatment for various times, the activity of DNA synthesis in the repopulating pachytene spermatocytes is obviously lower than that in the normal control,the synthetic activity of DNA is reduced by l4%,26%,40%,42%and 40%from the rats received gossypol treatment for 3,4,6,7 and 8 weeks,respectively.
