低温冻存人外周血单个核细胞的核酸、酶与糖原活性研究

摘  要:用定性定量组织细胞化学方法,对正常及个同降温条件冻存的人外周血单个核细胞的DNA、碱性磷酸酶、过氧化物酶、琥珀酸脱氢酶活性及糖原含量进行测定.在程序降温过程中,未消除样品融合热释放的人外周血单个核细胞的DNA、碱性磷酸酶、琥珀酸脱氢酶、过氧化物酶活性及糖原含量均显著下降.消除融合热后,冻存人外周血单个核细胞DNA活性变化不显、碱性磷酸酶、琥珀酸脱氢酶活性及糖原含量略有下降,过氧化物酶活性略增加.说明人外周血单个核细胞低温冻存时,在降温过程中消除融合热的释放是保持单个核细胞生物活性的重要环节.Activity of dexoribonucleic acid(DNA), Alkaline phosphatase (AKP), Succinate dehydrogenase (SDS), Peroxidase(POX), and the amount of Glycogen in human peripheral blood mononucleated cells frozen at different low temperature were measured by qualititative and quantitative histochemistry methods. The results were as follows: The activity of DNA, SDS, AKP, POX and the amount of Glycogen in human peripheral blood mononucleated cells with fusion heat reduced remakably. After fusion heat in the freezing cells was released, the activity of DNA in cryopreserved human peripheral mononucleated cells remained the same, the activity of SDS, AKP and the amount of Glycogen in the mononucleated cells reduced slightly but the activity of POX in these cells increased slightly. The results showed that the release of fusion heat from human peripheral mononucleated cells suspension when frozen at low temperature was an important factor in maintaining the biological activity of human peripheral blood mononucleated cells.
