摘 要:胚胎大鼠脊髓植块接种于纯度>95%的单层人胎儿雪旺氏细胞表面进行培养,观察雪旺氏细胞对神经突起生长的支持作用.脊髓神经突起在雪旺氏细胞表面活跃生长,突起生长速度>450μm/天,与对照组相比有显著性差异(P<0.05).将长于鼠尾胶上的雪旺氏细胞移植于成年大鼠脊髓(T10~12)损伤腔内,移植后21天后,透射电镜观察,有髓或无髓纤维长入移植物中,移植后30天,长入移植物中的轴突更丰富,而无细胞的胶原移植物中未见轴突长入.结果表明,雪旺氏细胞在体内、外能支持和促进中枢神经轴突生长.Several pieces of CNS tissue from embryonic rat spinal cord were explanted onto the monolayer of human fetal Schwann cells to examine the ability of Schwann cells to support and promote the neurite outgrowth of CNS. Neurites grew vigovously and singly or in fine fasciles on Schwann cells at the rates of more than 450 μm a day, however neurites grew slowly on acellular collagen. Polymerized collagen rolls containing human fetal Schwann cells incubated in culture were grafted into the lesioned cystic cavities of the thoracie spinal cord of adult rats. Axons were present within the graft 21 days after the implantation and both ensheathed and myelinated axons became numerous 30 days after implantation. No axonal ingrowth was found in acellular collagen rolls. The result suggests that Schwann cells can support and promote the axonal growth of the central nervous system both in vitro and in vivo.
