白三烯受体基因mRNA表达水平与儿童哮喘的相关性

摘  要:目的:检测了白三烯受体基因mRNA在儿童外周血白细胞中的表达;研究轻、中、重度哮喘患者白三烯受体基因mRNA表达水平是否存在不同;探讨性别和年龄因素对哮喘组白三烯受体基因mRNA表达水平产生的影响。方法:应用逆转录聚合酶链式反应(RT-PCR)技术,对比并定量分析CysLT1-Receptor和CysLT2-Receptor基因mRNA在哮喘组和对照组之间的表达差异。结果:哮喘组及对照组中均有CysLT1-Receptor和CysLT2-Receptor基因mRNA的表达;哮喘组白三烯受体基因mRNA的表达显著增高,(P均<0·05);轻、中、重度哮喘患儿CysLT1-Receptor和CysLT2-Receptor基本mRNA表达水平无显著性差异;性别、年龄因素对哮喘组白三烯受体基因mRNA表达无明显影响P>0·05。结论:CysLT1-Receptor和CysLT2-Receptor基因在儿童外周血白细胞中存在表达,其mRNA表达水平在哮喘组显著增高。性别和年龄因素对哮喘组白三烯受体基因mRNA的表达无明显影响。Objective: To explore gene expression of eysteinyl leukotrienes (CysLT) receptor gene mRNA in peripheral blood of children and its difference among mild, moderate and severe asthma patients and its influential factors, including sex and age. Methods: Expression difference in CysLT1 - receptor and CysLT2 - receptor gene mRNA between asthma group and control group was quantificationally analyzed by RT- PCR. Results: There were CysLT1 -receptor and CysLT2 -receptor gene mRNA expressions in asthma group and control group ; CysLT receptor gene mRNA in asthma group significantly increased ( P 〈 0. 05 ), there was no significant difference in CysLT1 - receptor and CysLT2 - receptor gene mRNA among mild, moderate and severe asthma patients. Sex and age were not related to CysLT receptor gene mRNA expression ( P 〉 0. 05 ) . Conclusion: CysLT1 - receptor and CysLT2 - receptor gene express in children's peripheral leucocytes; Rhe levels of CysLT1 - receptor and CysLT2 - receptor mRNA expressions increase in asthma group. Sex and age have no obvious effect on CysLT receptor gene mRNA expressions.
