摘  要:目的探讨AT5BIVA和GM0639成纤维细胞系在体外培养条件下的增殖、O(—·)2的释放情况及佛波醇肉豆蔻醋酸酯(PMA)和二亚苯基碘(DPI)对两种细胞系增殖及O(—·)2释放量的影响.方法两种细胞系分别培养24、48 h,以3H-TdR参入法分别测定GM0639和AT5BIVA两种细胞系cpm值,了解两种细胞系的增殖情况,以细胞色素C还原法检测两种细胞系培养介质中O(—·)2浓度.同时观察PMA及DPI对两种细胞系增殖和O(—·)2释放的影响.结果两种细胞系增殖过程中自身能够释放O(—·)2,PMA可增加两种细胞系O(—·)2释放量,促进二者增殖;而DPI减少两种细胞系O(—·)2释放量,抑制二者增殖.结论低水平O(—·)2促进细胞增殖,且AT5BIVA细胞增殖速率随O(—·)2浓度变化而变化的幅度大于GM0639细胞,提示ATM基因突变是引发这一现象的可能原因.Objective To study the role of superoxide anion free radical (O2 ) in promoting proliferation of fibroblast cell lines ATSBIVA and GM0639. Methods Fibroblasts were cultured with RPMI1640 medium containing 10% neonatal calf serum at 37℃ in 5% CO2 incubator, stimulated directly by phorbol 12-myristate 13-acetate (PMA) and diphenylene iodonium (DPI). The concentration of O2 was measured by cytochrome C reduction. The effect of O2 in promoting proliferation of fibroblasts was investigated by ^3H-TdR incorporation liquid scintillation counting. Results The proliferation of AT5BIVA and GM0639 fibroblasts was induced obviously by low concentration of O2. PMA could promote the proliferation of two kinds of fibroblasts by increasing the generation of O2, and DPI could inhibit the proliferation of two kinds of fibroblasts by decreasing the generation of O2. Conclusion The increase of low concentration of O2 is closely related to the proliferation of two kinds of fibroblasts, and the proliferation rate of AT5BIVA fibroblast alters more quickly than GM0639. One with the change of the concentration of O2. ATM gene mutation may be responsible for it.
