摘 要:目的 研究毒乃清颗粒对模型大鼠肾脏转化生长因子β1(TGF-β1)表达的影响,初步探讨其作用机理。 方法 采用改良法对大鼠进行5/6肾切除以及腹腔注射链脲佐菌素制作糖尿病肾病(DN)肾功能不全模型,设立健 康组、假手术组、病理组、尿毒清组、毒乃清治疗组进行对照,采用免疫组化方法观察干预后各组大鼠肾脏TGF- β1表达情况(计算机图像半定量分析)。结果 病理组大鼠肾脏TGF-β1的表达明显高于健康组及假手术组,而毒 乃清治疗组TGF-β1在肾脏中的表达明显低于病理组与尿毒清组,其染色面积与积分光密度均较病理组小 (P<0.01),与尿毒清组比较均更低(P<0.05)。结论 TGF-β1与DN的发生发展关系密切,毒乃清颗粒能够降低TGF- β1在DN大鼠肾脏中的表达,可能是毒乃清治疗DN的机理之一。Objective To study the effect of the later-phase diabetic nephropathy rats treated with Dunaiqing Granules on TGF-β1 expression in kidney. Methods Improved 5/6 nephrectomy and STZ abdominal injection to creat rat models of later-phase DN. The model rats were randomly divided into three groups, DN control, treated with Niaoduqing and Dunaiqing, as controlled by two groups, normal control, pseudo-operation control. Immunohisto-chemistry was used for detected expression of transforming growth factor β1 in kidney. Results After six weeks with Dunaiqing Granule treatment, administration of Dunaiqing Granules in the DN rats can down-regulate TGF-β1 expression in kidney, getting close to normal control. But it was up-regulated in DN control and Dunaiqing treatment group. Conclusions TGF-β1 expression was down regulated in the DN rats renal. This is probably related to DN progress. Down-regulated TGF-β1 expression might be involved in the therapeutical effect.
