摘 要:目的:考察植物生长物质对四裂红景天愈伤组织诱导和培养的影响并分析愈伤组织中红景天苷含量。方法:运用正交设计实验,在MS固体培养基中添加不同种类、不同含量的植物生长物质,进行愈伤组织的诱导和培养,并用HPLC检测愈伤组织中红景天苷的含量。结果:培养基中含有2,4-D1mg·L-1,NAA2mg·L-1,6-BA0.5mg·L-1,KT0.1mg·L-1时,愈伤组织诱导效果最好,诱导率为83.3%。当生长物质配比为2,4-D1mg·L-1,6-BA0.1mg·L-1和KT0.5mg·L-1时,最适于愈伤组织的培养,培养30d后干重达11.77g·L-1,红景天苷的含量为干重的0.28%。结论:红景天愈伤组织在诱导和培养2个阶段所需的生长物质种类和含量有很大差异;诱导的愈伤组织能够合成红景天苷。Objective: To investigate the effect of plant growth substances on induction and culture of callus from Rhodiola quadrifida and also to analyze salidroside contents in the callus. Method: The optimum combination of plant growth substances in MS solid medium for induction and culture of callus was established using orthogonal design. The contents of salidroside was analyzed by HPLC. Result: MS medi- um containing 2,4-D 1 mg·L^-1, NAA 2 mg·L^-1, 6-BA 0.5 mg·L^-1 mand KT 0.1 mg·L^-1 could induce the callus from R. quadrifida most effectively; the induction rate was 83.3%. The optimized combination of plant growth substances for callus subculture was 2,4-D 1 mg·L^-1, 6-BA 0.1mg·L^-1 and KT 0.5 mg·L^-1. The dry weight could reach 11.77g·L^-1 when the callus was cultured in the optimum medium for 30 d and salidroside content was 0.28%. Condusion: The quantities of plant growth substances required for induction and culture of callus axe different in R. quadrifida. The callus could produce salidroside.
