摘  要:从北京地区大白菜病株上分离到芜菁花叶病毒一个分离物(TuMvB),这个分离物侵染大白菜、油青菜、甘蓝,产生花叶症状,在心叶烟上表现系统枯斑,三生烟和千日红上产生局部枯斑。该分离物为线条状病毒,粒体大小为700—700hm×13—15nm,PEG沉淀后经二次差速离心得到的病毒纯化制剂,对紫外钱呈典型的核蛋白吸收,A260/A280为1.21;分析超离心获得病毒的沉降系数为145s.SDS-聚丙烯酰胺凝胶电泳测得:外壳蛋白亚基由单一组分组成,分子量为27,000道尔顿,病毒制剂在冰箱内存放2个月后电泳,产生两条蛋白带,分子量分别为27,000和25,000道尔顿。聚丙烯酰胺凝胶电泳测得:TuMVB-RNA的分子量为3.2×10~6遭尔顿。电镜下病组织超薄切片中存在风轮状内含体,还有环状体和带状体。以纯化病毒制剂为抗原制备了TuMVB的抗血洁,效价为1:1024。用酶联免疫吸附技术ELISA检测了田间大白菜上芜菁花叶病毒的带毒率为75.4%。The present paper deals with the further studies on one of the isolates of Turnip mosaic virus(TuMV_B).The Purified virus from infected leaves of Brassica juncea showed the following charateristics;A_(260/280)=1.21,a,ingle symmetrical absorbent peak appeared an analytical ltracentrifugation,s_(20)=145s,polyacr- ylamide gel electrophoresis in the presence of SDS showed that the subunit ofcoat protein is of a single component with a molecular weight of about 27,000Dalton.when the stored TuMV_B had been stored for two months,the coat pro-tein dissciated into two components,with molecular weight 25,000 and 27,000respectivly.The molecular weight of TuMV_B-RNA was estimatedfrom its mobi-lity during electrophoresis to be about 3.2×10~6 Dalton.Ultrasections of the infected leaves showed the appearance of a number of inclusion bodies inclu-ding pinwheels,rings and dense bands.Antiserum prepared from purified TuM-V_B resulted in a titre of 1/1024.During the Chinese cabbage growing season in 1986,samples of Chinese cabbage plants were collected from fields and detected with ELISA.It was shown that 75.4% of the samples tested containing TuMV_B. Now a faculty of Tianjin Academy of Agric.Science.
