速率法检测尿中N－乙酰－β－D－氨基葡萄糖苷酶

摘　　要：用新色原氯硝氨基苯基葡萄糖苷（ＣＮＰ－ＮＡＧ）速率法测定尿ＮＡＧ．尿ＮＡＧ测定的最适ｐＨ４．４～５．０．最佳呈色反应的ｐＨ４．６～５．０，在４００ｓ观察期内吸光度变化与时间成止比例．ε＝３．７９×１０３Ｌ·ｍｏｌ－１·ｃｍ－１，Ｋｍ＝０．５ｌｍｍｏｌ·Ｌ－１．ＴｒｉｎｔｏｎＸ－１００可促进底物的溶解．尿ＮＡＧ正常参考范围：１．２５～１１．３３Ｕ／ｇＣｒｅ．new chromogenic substrute 2-choro-4-nitrophenyl-N-acetyl-β-D-glucosaminide,and kinetic methed ware wsed to determine the activity ofN-acetyl-β-glucosaminidase (EC 3·2·1· 30;NAG)in urine.Chromogen CNPdeveloped yellow color in pH4,6～5.0,and the absorbance change during enzymatichydrolyzations was measured automatically.The Km was 0.5l mmol·L-1,ε was 3.79 ×103 L·mol-1·cm-1. The reference of urinary NAG was≤l.25～11.33 unit per gram ofcreatinine(n = 152 )
