尿中半乳糖苷酶活力的连续监测法

摘　　要：本文报道用新色原氯硝基苯基Ｎ－乙酰－β－Ｄ－氨基半乳糖苷（ＣＮＰ－ＧＡＬ）连续监测法测定尿ＧＡＬ。此法不需尿样空白，有很高的精度，批内、批间的ＣＶ≤６．６８％。尿ＧＡＬ的最适ｐＨ４．４～５．０，最佳呈色反应ｐＨ４．６～５．０，在４００秒观察期内吸光度变化与时间成正比例。ε＝３．７９×１０~３Ｌ·ｍｏｌ^（－１）·ｃｍ^（－１），Ｋｍ＝０．３７ｍｏｌ／Ｌ。ＴｒｉｔｏｎＸ－１００可促进底物的溶解。葡萄糖、ＶｉｔＣ、胆红素对本法无干扰。尿ＧＡＬ正常参考范围：０．６９～６．７０Ｕ／ｇＣｒ（ｎ＝１５８）。此方法适用于各种自动化分析仪，可用于临床肾病的诊断。In this paper.we report to use a new chromogenic substrute 2-choro-4-nitrophenyl-N-acetyl-β-D-galactosaminide.and kinetic methed for determining the activity of N-aectyl-β-D-galaetosidase(EC 3.2.1.23.GAL) in urine by autoanalyzer.No sample blank is required. Very high precision is possible.with CV of≤ 6.68% both within-run and between-run. Chromogen CNP developed yellow color in pH 4.6-5.0.and the absorbance change during enzymatic hydrolyeations was measured automatically.The Km was 0.37 mmol/L.ε was 3.79×10￣3 L.mol￣(-1).cm￣(-1).Glucose.ascorbic acid. bilirubin do not interfere.The reference value of urinary GAL was 0.69-7.59 unite per gram of creatinine.The mothed is suitable for use with various autoanalyzers.and it should usefulfor diagnosis renal diseases.
