摘 要:在高(15mmol·L(-1))条件下.野生型粪产碱菌A1501远离水稻根表,工程菌A1513(nifA)和A1523(ntrC-nifA)仍能聚集在报表的粘质层。测定果胶酶活性表明,在高条件下,工程菌比野生型有较高的果胶酶活性,说明果胶酶在联合过程中可能起重要作用。根系粘质与粪产碱菌及其工程菌的胞外多糖(EPS)相互作用的热力学研究表明:EPS与根系粘质的结合存在放热和吸热过程,野生型A1501和工程菌A1513均表现为放热吸热再放热再吸热的过程,而工程菌A1523其EPS与根系粘质反应只有放热反应,没有吸热过程。The attachment of Alcaligenes faecalis to the host plant rice was observed by Laser confo-cal Micrographs (LCM),it was found that the nifA (A1513)and ntrC-nifA (A1523) transconjugants ofA. faecalis can be colonized on rice root surface under the high concentration of ammonium-containing cul-tural medium (15mmol/L(-1)),while the wild type(A1501)was removed from the rice root surface underthe same condition. The reason is remained to exploration, but the peclolytic activities were still remainedfor A1513 and A1523 under this condition, suggesting that the pectolytic activity might be some roles inthis attachment. The experiment was also carriedout by litration of exopolysaccharides (EPS) from differ-ent mutants with rice root mucigel (RRM) and monitored by calorimeter with isothermal unit. The resultsshew that there were exothermic process and followed by endothermic process for A1501 and A1513, butonly exothermic process for A1523. These processes indicated that the exothermic process occurred at thephysical stage, it might be the adsorption process. and the endothermic process is a chemical stage,itmight appeared at the anchoring process. It is remained to further investigation.
