摘　　要：用活化的抗癌药物氨甲喋呤（ＭＴＸ）和羊抗鼠ＩｇＧ（ＳＡＭＩｇＧ）共价结合，制备ＳＡＭＩｇＧ－ＭＴＸ结合物，通过ＣＥＡ单抗（ＭｃＡｂ）的介导，ＳＡＭＩｇＧ－ＭＴＸ能选择性地杀伤ＣＥＡ＋细胞，其选择细胞毒作用是ＣＥＡＭｃＡｂ－ＭＴＸ结合物的２倍，提示间接导向治疗具有简便、经济、高效的特点。Methotrexate actived with active ester was combined with sheep anti-mouse IgG. Through the mediation of anti-CEA monoclonal antibody, sheep anti-mouse IgG-methotrexate conjugates killed the human colon carcinoma cell line HT-29 (CEA positive) selectively. The cytotoxicity of indirect targeting was two times of that of direct targeting.This study showed that the indirect target was more simple, economic and effective than direct targeting and may be used for tumor biotherapy.
