淋巴细胞合成生长激素对自身细胞增殖及γ-干扰素产生的影响

摘  要:淋巴细胞能够合成和分泌生长激素已被证实。本文的目的在于观察免疫反应性生长激素(irGH)在体外诱导淋巴细胞应答反应中发挥的作用。以特异性抗体和irGH结合进行阻断试验,结果显示PHA诱导的淋巴细胞增殖程度显著下降。45μg/ml的免抗人GH抗体使淋巴细胞增殖的抑制率达41.2%;使γ-干扰素(IFN-γ)的产生从56.3±27.4ng/ml下降到49.1±23.0ng/ml(P<0.05)由此说明irGH参与体外淋巴细胞应答活动的调节。It has been shown that lymphocytes produce immunoreactive GH (irGH). The activity of irGH in lymphocyte proliferation and IFN-gamma production was investigated with blocking test by anti-GH antibody in this study. The results showed that rat anti-human GH antibody (RAHGH) potently inhibited PHA-induced lymphoproliferation in serum free medium with a dose-dependent manner(r=0.984).Cells with RAHGH (45μgm/ml) had less (41.2%) incorporation of 3H-TdR in PHA-stimulated lymphocyte culture. Nonspecific antibody (RAMIg 50μg/ml) had no such effect. Neither RAHGH nor RAMIg affected the viability of cultured lymphocytes.In addition, treatment of RAHGH significantly decreased the level of Con-A induced IFN-gamma (from 56.3±27.4μg/ml down to 49.±23.0μg/ml,P<0.05), Collectively, these data indicated that lymphocyte-derived GH may play a critical role in replication and cytokine production of lymphocytes by autocrine/paracrine way.
