摘 要:利用~8H-Prazosin结合试验和不可逆阻断剂N-ethoxycarboxyl-2-ethoxy-1,2-dihydroquinoline(EEDQ)测定了老年大鼠大脑皮质α_1-肾上腺素受体(α_1-AR)含量及α_1-AdR的受体动力学变化。老年大鼠大脑皮质α_1-AdR含量明显低于青年;老年与青年大鼠大脑皮质α_1-AdR的半恢复时间(T_(1/2))分别为107.8h和64.1h,老年大鼠大脑皮质α_1-AdR的生成速率和降解速率常数分别为0.790pmol/(h·g蛋白)和6.43×10^(-3)/h,青年大鼠则分别为1.928pmol/(h·g蛋白)和0.0108/h。结果表明老年大鼠大脑皮质α_1-AdR更新缓慢。By using 3H-pra/osin binding assay and N-ethoxycarboxyl-2-ethoxy-l,2-dihydroquinoline (EEDQ), the densities of alpha-1-adrenoceptor and the receptor kinetics constansts of aged rats were measured. The alpha-1-adrenoceptor density of aged rats was lower than that of young rats (P<0.05). The 50% recovery times-of aged and young rats were 107.8h and 64.Ih; the reappearance rates of both rats were 0.790 pmol(h-g protein) and 1.928pmol (h-g protein); The constants of degradation of aged and yeung rats were 6.48×10-3/h and 0.0108/h respectively Our results showed that the recovery of alpha-1-adrenoceptor was slowed during aging.
