摘 要:在成功地制备了具有谷胱甘肽过氧化物酶(GPX)活性含硒抗体酶(Se-abzyme)的基础上,我们筛选了制备Se-abzyme的最佳条件,并对其理化性质及酶学性质和稳定性进行了深入的研究。结果表明,Se-abzyme的等电点为6.95和7.08,分子量为158kd;最适pH和最适温度范围比天然酶宽广;抗体酶的贮藏稳定性比天然酶高。用X射线光电子能谱技术测得在Se-abzyme中含硒量为5molSe/mol。用酶联免疫法测得Se-abzyme对谷胱甘肽(GSH)的结合常数为1.11×107(mol/L)-1。二硫代双硝基苯甲酸(DTNB)可抑制Se-abzyme,抑制常数为9×10-2(mol/L)。We successfully prepared the Se-containing abzyme with GPX activity and further studied its physicochemical and enzymci properties and stabilities. Data showed that the isoelectric point of the abzyme was 6.95 and 7.08,and its molecular weight was 158kd.The ranges of optimum pH and temperature of the Se-containing abzyme were wider than those of the native GPX. The stability of the abzyme in storage was higher than that of GPX. The content of Se in the abzyme was found to be 5 mol Se/mol abzyme by X-ray photoelectron spectrum and the binding constant was 1.11×107M-1 as assayed by ELISA method.The Seabzyme was inhibited by dithiobis (2-nitrobenzoic acid),and inhibition constant was determined to be 9×10-2M-1.
