摘 要:本文对大鼠在正常动情期和早期妊娠阶段(d1~d10)外周血雌二醇(E2)水平和子宫中雌二醇受体(ER)基因的表达(以3H-E2配体结合饱和分析测定胞浆受体ERc和核受体ERn,以Northern印迹法测定ERmRNA)作了全面的分析。观察到:(1)在正常动情期和妊娠早期,子宫ER基因表达受到E2的上调作用,而且ERmRNA和蛋白水平基本同步,说明这种影响是在基因转录水平;(2)受孕后,ER基因表达水平随外周血E2的升高也逐步上升,于d4~d5(着床前夕)达到高峰,d6~d7(着床中)突然下跌至低谷,随后回升;(3)孕后d6~d10子宫着床部位的ER表达明显低于非着床部位,同时胚胎也有一定的ER表达。The correlation of serum estradiol concentration and uterine estrogen receptor (ER) gene expression (ERn and ERc quantitated by Dextran Coat Charcoal assay and mRNA by Northern blotting) was studied during the rat estrous cycle and early pregnant stage (d1~d10). The ER geneexpression was up-regulated by estrogen and the levels of ER mRNA synchronized with thechanges of ER protein,suggesting that estrogen influcenced the transcriptional step of the ERgene. Postcoitum ER expression increased with the serum estradiol progressively,reached a peakon d4~d5 (just before implantation), but drastically dropped to the nadir on d6~d7 (during implantation) and then recovered. It was of interest to discover that ER mRNA level in the non implantational sites (NIS) of uterus was much higher than that in the implantational sites (IS).
