摘 要:本文报告用气相色谱仪测定利多卡因血浓度的一种新方法和在正常人中利多卡因药代动力学的研究。本测定法以1%XE-60填充的玻璃柱及氢火焰离子化检测器进行。柱温210℃,进样口温度270℃,载气N_2的流速40ml/min。利多卡因和内标毗咯卡因的保留时间分别为1.64min和2.45min。本法准确、灵敏、快速,最低检测浓度为0.05μg/ml。适用于利多卡因药代动力学研究和血药浓度监测。健康志愿者5人,单次静注利多卡因1mg/kg后的药时曲线经计算机以非线性最小二乘法自动拟合,符合二室开放模型。平均药时方程是C_t=1.64e^(-0.1473t)+0.51e^(-0.008t),主要动力学参数为t1/2α=5.4±2.0min,t1/2β=88.9±16.6min,V_c=0.544±0.291L/kg,V_d=1.709±0.411L/kg,CL_B=13.3±2.2ml/kg/min。The paper reported a new method for determination of bloodconcentration of lidocaine with gas chromatography and pharmacokineticstudy of lidocaine in healthy volunteers. A U-Shaped glass tube packedwith 1% XE-60 on chromosorb W (60-80mesh) and a flame-ionization detec-tor were used in the gas chromatographic analysis. Operating conditionswere: oven temperature 210℃, detector and injector port 270℃, nitrogen 40ml/min, hydrogen 30 ml/min, air 800 ml/min. Retention time of lidocaineand internal standard pyrrocaine were 1. 64 min 2. 45 min respectively. Themethod was precise. sensitive ane rapid. The minimum detectable concentra- tion was 0. 05μg/ml. The method can be used for pharmacokinetic study andtherapeutic concentration monitor of lidocaine. The parmacokinetics of lidocaine was studied in 5 healthy volunteersafter a single intravenous injection 1 mg/kg. The data was fitted by thenonlinear regression automatic program and showed to be subjected to twocompartment open model. Tae plasma concentration-time equation was C_t=1 .64e^(-0.1473t)+0.51e^(-0. 008t).
