摘 要:本文利用免疫沉淀、双相凝胶电泳技术分析了某些DR位点纯合子细胞DR抗原电泳图谱。待检细胞用~3H-亮氨酸生物合成性标记,提取膜蛋白,对抗DR单态型抗血清做免疫沉淀,沉淀物用双相凝胶电泳进行分析。结果表明,DR抗原α肽链电泳图单一,各DR型间无差异:β肽链电泳图谱因DR型别而异,是区分DR型别的主要依据。此外,2例DR2纯合子细胞图谱明显不同,提示为两个不同的亚型。In the present study the molecular structure of HLA-DR antigens from a panel of DR homozygous cells with DR1, DR2, DR7 and DR 9 specificity were analysed by 2-D (NEPHGE/SDS-PAGE) gel electrophoresis. The results demonstrated that all immunopreci pitates were composed of one a chain spot of MW35000 daltons at the isoelectr ic point (pI) of 5.0 and an invariant polypeptide (Ii) of MW 31000 daltons at pI 7.0.However, the structural differences of the β chains among DR1, DR2, DR7 and DR9 antigens were clearly demonstrated. The DR 2 β chain could be easity distingui shed from others. In addition, the electrophoretic patterns of two DR2 β chains were slightly different. Thus the results indicated that there existed two distinct subtypes of DR2 alloantigens.
