反向间接血凝法检测对虾肝胰腺细小病毒的方法学研究

摘　　要：用密度梯度离心纯化的对虾肝胰腺细小病毒（ＨＰＶ）颗粒免疫家兔制备特异性抗血清，并建立了一种反向间接血凝法以诊断ＨＰＶ感染。该方法的敏感性可达ｎｇ／ｍｌ病毒蛋白的检测限水平。通过对１０份已固定的患病对虾肝胰腺样品分别取一小部分进行检测，证实反向间接血凝法与组织病理检查的相符性良好，而前者具有方法简便、重复性好、试剂稳定和样品检测可在１．５—２小时内完成等特点，适合在现场推广使用。It is described in this essay that the specific antiserum was prepared byimmunizing rabbit with the purified hepatopancreatic parvovirus (HPV ) and a reverse indirect hemagglutination(RIHA ) method for the diagnosis of the HPV infection was estab lished. The detecting sensitivity of the method is at the ng/ml level of viral proteins. Withthe detection of 10 small pieces of specimen from different infected hepatopancreas that hadbeen fixed in fixative. it shows that the RIHA was well in line with histopathologicalexamination. but the former is much simpler and more convenient to perform and repeatswell. Owing to that the reagent of the method is very stable and the detecting procedure canbe finished within 1. 5-2 hours, the method is suitable for application in .he field.
