摘 要:用Van De Water内耳器官培养系统或周祥宁和Van De Water建立的HEMA水凝胶作支持物的器官培养法,研究了鸡胚内耳原基的体外发育。这些内耳原基在体外培养12天后,有基底乳头(相当于哺乳动物的耳蜗),半规管的壶腹嵴、内淋巴腔和内耳软骨性包囊发育。基底乳头在内淋巴腔面有感觉毛细胞,其表面可见盖膜。超微结构研究证实内耳感受器有发育良好的毛细胞。内耳器官培养法不仅可用以探讨内耳发育和影响内耳发育的因素,研究神经营养因子对内耳感受器与位听神经节神经元发育的调控作用和对被损伤神经元修复的刺激作用,而且可提供实验模型,研究耳毒性药物,放射线和病毒对内耳的危害,以及内耳感受器毛细胞的再生,为感音神经性聋的预防和治疗铺平道路。The development of otocysts of the chicken embryo was studied m vitro,usmg HEM A hydrogel organ culture system developed by Zhou &. Van De Water or the organ culture system reported by Van De Water & Ruben. After 12-14 days' cultivation, basilar papilla in the lagena, semicircular ducts with their associated otic capsule and statoacoustic ganglion were observed. Sensory hair cells with overlymg tectorial membrane could be identified in the basilar papilla. Well developed hair cells in the labyrinthine sensory neuroepithelium were con-firmed at ultrastructural level. The technique of organ culture of the inner ear can be used to investigate the mechanism of the development of the inner ear and the factors regulating its development as well as effects oi neurotrophic factors of the development of the neurons of the statoacoustic ganglion and on the repair of these damaged neurons. Organ culture of the inner ear can provide experimental models for studying the effects of oto-toxic drugs,radiation and viruses on the sensory receptors of the inner ear. It is also a very useful approach to m-vestigating mechanism and triggering factors of hair cell regeneration of the avian and mammal for prevention and cure of sensorineural hearing loss.
