武汉地区正常汉族人载脂蛋白B基因区域DNA多态频率的研究

摘　　要：应用载脂蛋白Ｂ（ＡｐｏＢ）ｃＤＮＡ探针（ＬＢ１．５），检测了２１名武汉地区正常汉族人血脂个体的ＡｐｏＢ基因限制性内切酶ＭｓｐⅠ限制性片断长度多态性（ＲＦＬＰ）等位基因频率。结果显示：２１份标本共出现２．３５ｋｂ（Ｍ_１）和２．６０ｋｂ（Ｍ_２）２条杂交片段；突变型等位基因Ｍ_２位于ＡｐｏＢ基因３’端侧翼的高变区（ＨＶＲ）内，其基因频率为０．０２４，并具有种族差异。Ｍ_２基因的检出和基因频率的获得，为进一步揭示ＡｐｏＢ基因区域在脂类代谢中的作用提供了一个很好的遗传标记。We have investigated MSPI allele frequencies of restriction fragment length polytnorphism(RFLP)of the ApoB gene in 21 Chinese healthy subjects, with the ApoB cDNA probe(LB1.5).The result showed that 2.35 kb (M_1)and 2.60 kb(M_2)hybridization fragments were found in 21 samples. The rare allele (M_2)of the Msp I RFLP in 3 hypervariable region(HVR)resulted from an insertion sequences, with the frequency being 0.024. The results of the present study suggest that M_2 gene frequency varies with racial groups and ApoB Msp I polymorphism may be a useful genetic marker in Chinese population.
