摘 要:用3H-胸腺嘧啶核苷和3H-尿嘧啶核苷掺入法及流式细胞术,观察猪肺动脉缺氧内皮细胞条件培养液(HECCM)和1-(2,6-二甲基苯氧基)-2-(3,4-二甲氧基苯乙氨基)丙烷盐酸盐(DDPH)对猪肺动脉平滑肌细胞(PASMCs)DNA、蛋白质合成及细胞周期的影响。结果显示:HEC-CM可促进PASMCs的DNA合成,促进PASMCs由静止期(G0/G1)进入DNA合成期(S期)及有丝分裂期(G2+M期),但对其蛋白质合成无明显影响。DDPH对HECCM引起PASMCs的DNA合成增高的抑制率为59.71%,对G0/G1期细胞进入S期增高的抑制率为22.58%,此外,DDPH对PASMCs蛋白质的合成也有抑制作用,抑制率为24.93%。The effects of the medium conditioned by hypoxically cultured endothelial cells(HECCM)and 1-(2,6-dimethylphenoxy)-2-(3,4-dimethoxyphenylethy -lamino)propane hydrochloride(DDPH)on DNA, protein synthesis and cell cycle of the porcine pulmonary arterial smooth muscle cells(PASMCs )were studied using  ̄3H-thymidine and  ̄3H-uridine incorporation and flow cytometry. The results showed that HECCM stimulated DNA synthesis of PASMCs and promoted PASMCs from static phase(G_0/G_1)to DNA synthetic phase (S)and mitotic phase(G_2+M), but it had no obvious effect on protein synthesis of PASMCs. The rate of inhibition of DDPH on increased DNA synthesis of PASMCs induced by HECCM was 59.71%.The rate of inhibition on increased PASMCs from G_0/G_1 phase to S phase was 22. 58%, In addition, DDPH could decrease the protein synthesis of PASMCs(24.93%).
