摘  要:室温下,诸葛菜(Orychophragmusviolaceus)花粉寿命较短,7天后全部失去生活力,在4℃低温下,花粉寿命延长至15天,未干燥的新鲜花粉不耐冰冻,干燥花粉在-20℃保存40天时,萌发率降至25%,在液氮(LN)中,干燥花粉生活力基本不变,经DMSO溶液预处理的冰冻花粉生活力很低,在-20℃,花粉游离脯氨酸(Pro)含量随储存时间延长微弱下降.在LN中,花粉游离Pro含量不随时间变化。与新鲜花粉相比,LN保存花粉的游离Pro含量略有升高,但不显著,冰冻(-20℃,LN)保存20天的花粉均能正常授粉结实。Pollen preservation of O.violaceus was studied preliminarily.The result showed that Pollen life span wus shorter al room temperatute.Pollen viability was fort completely in 7 days at room remnperature or in 15 days at 4℃.Non dried Pollens could not risist to frost. During a period of 40 days.the survival Percentage of dried pollens stored at-20℃ declined to 25%.whereas the viability of dried pollens stored in liquid nitrogen(LN) remained constant.The viabillty of frozen pollens ptetrealed with DMSO solution was very low.As time went out,the content of free Pro in pollens declined slightly at-20℃ and did not change in LN.Compared with fresh Pollens.the content of free Pro in Pollens stored in LN rose up a little,but the difference was not remarkable. Pollens.stored respectively at-20℃and in LN for 20 days.could be used to pollinate and the pollinated flowers developed fruit and seeds.
