摘  要:本文采用显微镜观察和逐个细胞突起长度测量及MTT等方法研究了添加肽对无血清培养的C_6细胞生长的影响:在培养早期(9—36h),10^(-8)mol/L的AVP、NLPR或ZNC(C)PR等都能刺激生长;同浓度的OXT或ZDC(C)PR在起始时(9—17h)无明显影响,但稍后(36h)显示抑制作用。MTT染色法分析的结果指出:神经肽促生长作用主要表现在细胞生长前期(17h)并且是肽浓度依赖的,ZNC(C)PR的最低有效剂量10^(-10)mol/L比AVP的要低百倍。在控制条件下细胞缓慢生长时:上述肽内仅ZNC(C)PR和NLPR两者(10^(-8)mol/L)对C_6细胞的突起发生仍保留明显的促进作用。The effects of synthetic peptides on the growth of C6 cells in the serum-free culture were microscopically observed and quantitatively measured, and spectrodensitometrically determined by means of MTT staining. In early stage of incubation; (9-36 h),10-8 mol/L arginine-vaso-pressin (AVP), NLPR or ZNC(C)PR markedly stimulated cell growth while oxytocin (OXT) or ZDC(C)PR showed a significant inhibition at h-36. Results generated from MTT experiments confirmed that the stimulating effect of these peptides mainly appeared in the early stage of cell growth (9-17h) and were concentration-dependent. The lowest effective dose of ZNC (C) PR (10-10mol/L) was hundreds times lower than AVP. When C6 cells were in a conditioning growth situation, both ZNC(C)PR and NLPR (10-8mol/L) still markedly showed a facilitation on the neurites elongation.
