摘 要:采用小鼠子宫颈癌14号(U_14)给615近交系小鼠后肢肌肉内移植建立腹膜后淋巴结和肺的双向转移模型,将小鼠随机分为3组。脂质体组每次给子腹腔注射卡铂多相脂质体0.5mg/0.5ml;水剂组每次腹腔注射卡铂水剂0.5mg/0.5ml;对照组每次仅注射生理盐水0.5ml,每组隔日注射。肿瘤移植后第14~17天,对照组均发生腹膜后淋巴结转移和92.31%的肺转移,水剂组淋巴结转移率为90.91%,肺转移率为63.64%;而脂质体组的转移率最低,分别为70%和40%。对照组淋巴结和肺几乎全是“++^+++”转移,而脂质体组淋巴结在14~15天仅为“±^+”转移,第17天时转移级别上升,肺的转移级别以“±^+”为主。水剂组淋巴结和肺的转移率和转移级别介于脂质体组与对照组之间。脂质体组的下肢移植瘤明显小于水剂组和对照组。提示,卡铂多相脂质体腹膜注射对小鼠移植瘤腹膜后淋巴结和肺的转移具有明显地抑制或导向治疗作用,而卡铂水剂型作用欠佳。The uterine cervix carcinoma cells(U14)of mice were inoculated into the muscle of right hindlegs of inbred 615 strain micefor establishing a lymphnodes and lung, metastatic model. Then the mice were divided into three groups at random. 0.5mg/0.5mlcarboplatin polyphase liposome was injected into peritoneal cavity of each mouse of the liposome group once in two days for five times at all. 0.5mg/0.5ml carboplatin aqueous solution was injected for the aqueous solution group with the same method as the liposome group did. Normal saline solution was used only for the control group. From the 14th to the 17th day after inoculation, the metastasis rate of the retro-peritoneal lymphnodes in the control group was 100% and that of lung was 92.31%. The metastasis rate of lymphnodes in the aqueous solution group was 90.91% and the metastasis rate of lung was 63.64%,the polyphase liposome group obtained a lowest metastasis rate of lymphnodes (70%) and lung ( 40% ) .The metastasis degree ()-() was found in almost all lymphnodes and lung of the control group. But (±)-( + ) degree was found in most lymph-nodes of the carboplatin liposome group at the 14th or the 15th day and in lung from the 14th to the 17th day. Either metastasis rate or metastasis degree, the differences was obvious betweenthe liposome group and the con-trol group. But there were no significant difference between the aqueous solution group and the control group. The results of this study indicate that the car-boplatin encapsulated in polyphase liposome is superior to the carboplatin aqueous solution to suppress the metastasis of tumor cell ( U14 ) to the retroperitoneal lymphnodes and the lung of mice.
