摘  要:目的:探讨干扰素-γ、干扰素-α能否抑制玻璃体内细胞增生。方法:应用~3H-胸腺嘧啶核苷(~3H-TdR)掺入及液体闪烁测量技术,观察不同浓度的干扰素-γ、干扰素-α(10IU/ml~10~4IU/ml)在3天、5天两个时间点对体外培养的人视网膜色素上皮细胞的影响。结果:在3天时观察,干扰素-γ对人视网膜色素上皮细胞有轻度的抑制作用,最大(10~4IU/ml)抑制率为26%(P<0.05),干扰素-α对视网膜色素上皮细胞有轻度的抑制作用,最大(10~4IU/ml)抑制率为26%(P<0.05)干扰素-α对视网膜色素上皮细胞的抑制作用,较干扰素-γ弱(10~4IU/ml时抑制率为16%)。5天时观察,抑制作用较3天时减弱。结论:干扰素-γ、干扰素-α对人视网膜色素上皮细胞有轻度的抑制作用,其作用维持3天左右。PURPOSE. To evaluate the effects of interfe-ron-gamma (IFN-γ) and interferon-alpha (IFN-α) on human retinal pigment epithelial (RPE) ceils in vitro. METHODS: Cultures of human RPE cells were incubated with either of the IFNs (10IU/ml~10~4 IU/ml) for 3~5 days, and proliferation rates of the cells were assayed by [~3H]-thymidine incorporation and liquid scintillation techniques. RESULTS: There was a slight inhibition on human RPE cells with dosages from 10IU/ml to 10~4IU/ml. The inhibition rates of IFN-γ of 10~3IU/ml, 10~4IU/ml were 8%, 15%, 26%, and of IFN-α were 7%, 9%, and 16%, respectively. On the 5th day there was a slight decrease in the inhibition rate than on the 3rd day. CONCLUSION: IFN-γ and IFN-α exerted a slight inhibition on human RPE cells lasting 3 days.
