高效液相色谱法测定人血舒林酸及其代谢物浓度

摘  要:本实验采用高效液相色谱法测定人血中舒林酸及其代谢物(硫化物、砜)的浓度。色谱柱为SpherisorbC18(5μ,4.6mm×200mm);流动相为乙腈-4%冰乙酸(50:50)5min,乙腈-4%冰乙酸(80:20)15min;检测波长为328nm。采用吲哚美辛作为内标物,以峰高度定量。舒林酸、硫化物、砜的平均回收率分别为:(82.78±5.15)%、(74.19±2.93)%、(85.88±4.91)%。日内及日间RSD均<10%。本法适用于药代动力学和临床研究。A HPLC method for the determination of sulindac and its sulphone and sulphide metabolites in human serum was developed.The method utilized a Spherisorb C18 column,acetonitrile -0.4%acetic acid as mobile phase of an initial ratio of 50:50 for 5 min and then 80:20 for 15 min with a flow rate of 1ml/min,indomethacin as an internal standard.The absolute calibration curve(sulindac:C=0.5500H+0.0061,r=0.998,n=5:sulphone:C=.5760H+0.0836,r=0.9999,n=5;sulphide:C=0.4069H-0.0829,r=0.9999,n=5)was obtained with the range of 0.15~4. 8μg/ml.The average absolute recovery of sulindac,sulphone and sulphide from serum was(82.78±5.15)%,(74.19±2.93)%,(85. 88±4.91)%,respectively.The intraday and interday RSD were less than 10%.
