冻存增强人外周血单个核细胞产生IL-2的机理初探

摘  要:作者用ELISA法研究了冻存对人外周血革个核细胞(PBMC)产生白介素2(IL-2)的影响。结果表明,冻存后PBMC产生IL-2能力明显增强;去除CD8+细胞或加入消炎痛后,PBMC产生IL-2能力增强,且与PBMC冻存后产生IL-2水平相同。研究提示:冻存过程有可能通过抑制PGE2分泌细胞及使CD8+细胞失活,使PBMC增加IL-2的产生。To better understand the effect of cryopreservation on various immunocompetent cell functions,we have examined the interleukin-2 (IL-2) producing activities of frozen mononuclear cells (PBMC) from healthy subjects. The level of IL-2 in culture supernatants of PBMC was assayed by the ELISA method.The results showed that the frozen PBMC produced significantly larger quantities of IL-2than fresh cells. Similar to freezing,indomethacin treatment also increased the IL-2 producing activities of fresh cells. but could no longer enhance the production of IL-2 by frozen cells. Elimination of CD8+ T cells from PBMC also resulted in the increase of IL-2 by fresh cells and freezing did not further enhance the IL-2 secreting activities of PBMC, which were devoid of CD8+ T cells. These data indicate that inactivation of CD8+ T cells and inhibition of PGE2 secreting cells during the freezing procedure are responsible for the increase in IL-2 secretion.
