流行性出血热循环免疫复合物组分分析

摘　　要：应用ＳＤＳ－聚内烯酰胺凝胶电泳及免疫转印技术对流行性出血热患者血清中免疫复合物组分进行了分析。流行性出血热循环免疫复合物经ＳＤＳ－ＰＡＧＥ分离，考马斯亮兰染色，显色主要有７条带，分子量分别为２３ｋＤ、５０ｋＤ、５２ｋＤ、６５ｋＤ、７２ｋＤ、８０ｋＤ及１００ｋＤ。采用该病毒特异性抗血清、单克隆抗体以及人免疫球蛋白、补体成分抗血清识别，在其循环免疫复合物中可检出特异性病毒抗原及相应的免疫球状蛋白和补体成分。应用ＰＣＲ技术在部分患者循环免疫复合物中可检出病毒核酸。上述结果从分子水平证实流行性出血热患者的ＣＩＣ中存在病毒抗原与特异性抗体是其循环免疫复合物的主要组分，它们构成了不同类型的免疫复合物，并且大多数可以结合补体成分。DS-polyacrylamide gel electrophoresis and Western blot analysis were carried out on the components of circulating immune complex(CIC) in sera of patients with epidemic hemorrhagic fever(EHF).After separtion in SDS-PAGE,the cumponents of CIC were stained with coomassie brilliantblue showing mainly 7 bands with appwrant molecular weight about 23kD,50kD,52kD,65kD,72kD,80kD and 100kD,respoctively.EHF virus antigens,human lg and the component of complement couldbe detected in the CIC using the speci fic antibodies against EHF virus,human Ig and C_3,suggesting thatthe EHF virus antigen and human Ig are the main components of the CIC,EHF virus RNA was also detected In CIC from tome patients by RT-PCR,Furthermore,there were at least lgG,IgM,IgA type CICin the sera of EHF potients and most of them were combined with the component of complement.
