摘　　要：通过３Ｈ－ＴｄＲ掺入实验和ＲＮＡ印迹分析，观察了内皮素－１对大鼠血管平滑肌细胞ＤＮＡ合成及细胞增殖相关基因表达的形响。结果表明，内皮素－１作用于血管平滑肌细胞１２ｈ，３Ｈ－ＴｄＲ掺入开始增加，２４ｈ达到峰值。在内皮素－１作用下，原癌基因ｃ－ｆｏｓ和ｃ－ｊｕｎ进行快速瞬间表达，其表达活性在３０ｍｉｎ达到峰值，３ｈ后恢复到原来水平；Ｐ３４ｃｄｃ２激酶基因在内皮素－１刺激１２ｈ后表达活性开始增加，至２４ｈ仍维持在较高的水平。本文提示，内皮素－１对血管平滑肌细胞ＤＮＡ合成的诱导作用与促进细胞增殖相关基因表达有关。The effects of endothelin-1 on DNA synthesis and expression of genes related to cell proliferation in rat vascular smooth muscle cells (VSMC) were investigated by 3H-TdR incorporation and Northern blot analysis. The results suggest that 3H-TdR uptake increased at 12 h after stimulation with endothelin-1, with reaching a peak at 24 h.Endothelin-1 induced rapidly and transiently the expression of the proto-oncogenes c-fos and c-jun,the activity of their expression peaked at 30 min after stimulation, and rapidly declined there after. The expression of the p34cdc2 kinase gene augmented at 12 h after endothelin-1 stimulation and continuously expressed at relatively high levels until 24 h. These data indicate that endothelin-1 stimulates DNA synthesis of VSMC by inducing the transcription of genes related to cell proliferation.
