摘  要:本文研究了河南马氏钳蝎蝎毒(SVC)的三个分离毒素:SVCⅠ、SVCⅡ及SVCⅢ对Eca109细胞株的杀伤及生长抑制作用。结果表明,SVCⅠ和SVCⅡ对Eca109细胞有直接杀伤作用,它们对Eca109细胞的半数杀伤浓度(LC_(50))分别为7.999μg/ml、4.499μg/ml;SVCⅡ和SVCⅢ处理细胞96h后,对Eca109细胞的生长抑制率(IR%)分别达83.47%和92.29%;SVCⅡ、SVCⅢ对Eca109细胞的集落抑制率分别达81.3%、80.1%,它们对Eca109细胞的半数抑制浓度(IC_(50))分别为9.900μg/ml、3.099μg/ml;经SVCⅡ、Ⅲ作用后,Eca109细胞有丝分裂指数(MI%)亦明显减少;MTT法的实验结果显示,SVCⅡ、Ⅲ对Eca109细胞具有明显的细胞毒性作用。未观察到SVCⅠ对Eca109细胞的作用。The lethal and the growth-inhibiting effect of three components (SVC Ⅰ,SVCⅡ,SVCⅢ)isolated from scorpion venom on Eca109 cells were studied. The resultsshowed that SVC Ⅱ and SVC Ⅲ possess killing effect on Eca109 cells,the half lethalconcentration(LC_(50) )of the SVC Ⅱ and SVC Ⅲ on Eca109 cells were 7.999μg/ml and 4.499μg/ml respectively. Treated the cell with SVC Ⅱ or SVC Ⅲ for 96 hours,the growth inhi-biting-rate on Eca109 cells were 83.47% and 92.29% respectively. The colony-forminginhibiting rate of SVC Ⅱ and SVC Ⅲ on Eca109 cells were 81.3% and 80.1%,and the halfinhibiting-concentration(IC_(50))were 9.900μg/ml and 3.099μg/ml respectively. The mitoticindex(MI‰)of the Eca109 cells were decreased obviously after the cells were treatedby SVCⅡ and SVC Ⅲ. The cytotoxic effect of SVC Ⅱ and SVC Ⅲ on Eca109 cells was ob-served by means of MTT test.The effect of SVCⅠ on Eca109 cells was not found.
