Hyperplasia of myocardial collagen and its relation to myocardial angiotensin Ⅱ in rats during pressure and Volume overload

摘  要:对于机械负荷,尤其是容量负荷增加时,心肌胶原发生何种改变以及其机制如何,目前尚不十分清楚。本文验采用放射免疫及生化等方法,就大鼠动-静脉瘘(AVF)和主动脉狭窄(AC)时,心肌和血浆血管紧张素Ⅱ(AngⅡ)含量、心肌胶原总量(TCC)、含量(CC)及Ⅰ/Ⅲ型胶原比值的变化进行初步研究。发现在心肌AngⅡ明显升高的同时,AVF大鼠术后五周不仅心/体比值(V/BW)明显升高,其左右室TCC和CC也显著增加,但Ⅰ/Ⅲ比值无明显改变。AC大鼠从术后两周起,V/BW、左右室TCC、CC及Ⅰ/Ⅲ比值就显著高于对照组。而两种模型大鼠血浆AngⅡ未见升高。提示:1.容量超负荷持续一定时间后可导致心肌胶原明显增生,且Ⅰ和Ⅲ型胶原增生程度基本成正比例;2.心肌AngⅡ在两种负荷增加所致心肌胶原增生中可能起着重要作用。The mechanisms of the changes of heart collagens which were caused byhemodynamic overload,especially by volume-overload,still remained in suspending.Toapproach this end,the changes of ventricular and plasma angtotensin( Ang)Ⅱ content,total heart collagen content(TCC ),collagen concentration(CC)and the ratio of Ⅰ:Ⅲcollagen types(Ⅰ/Ⅲ)in aorta-constriction(AC)and aortacava fistula(ACF)ratsWere determined by means of radioimmunoassay and biochemical methods.It was foundthat not only the ratio of ventricle wet weight to body weight( V/BW)increased ob- viously,but also the TCC and CC in both left and right ventricles enhanced significan-tly,combined with the increase of ventricular AngⅡ content(whereas the Ⅰ/Ⅲ remainedunchanged ).In AC rats,associated with the increase of ventricular Ang Ⅱ,the V/BW,TCC,CC and Ⅰ/Ⅲincreased significantly two-weeks after the surgery·No enhan- cement of plasma AngⅡ in AC or ACF rats.These findings suggested that:1.Obvioushyperplasia of heart collagens occured after volume-overload and the hyperplasia of Ⅰand Ⅲ collagen types was generally in an hemogeneous fashion;2 .Local producedAng Ⅱmay play a cornerstone role in hyperplasia of heart collagens during pressureor volume overload.
