摘 要:应用聚合酶链反应(PCR)检查感染鼠疫的动物脏器标本并与细菌分离培养进行比较。在31份肝组织标本中PCR阳性11份(35.5%),细菌培养阳性9份(29.0%);31份脾组织标本中PCR阳性25份(80.7%),细菌培养阳性21份(67.7%)。对照组中,以假结核菌感染的小白鼠和未经感染的小白鼠脏器标本PCR全部阴性。结果表明,PCR法优于传统的细菌分离培养,具有快速、特异性强、敏感性高和不受杂菌污染影响等优点。适用于鼠疫自然疫源地动物鼠疫的监测。The PCR method was used to detect organ specimens of mice (containing liver samples and spleed samples )that were infected by Y. pest is and compared with the bacteria isolation and culture method. Of 31 samples of the liver tissues, 11 samples were positive by PCR(35. 5%),but 9 samples of those were positive by bacteria culture(29. 9%) ;of the 31 samples of spleed ,25 were positive by PCR(80. 7%),then 21 were positive by bacteria culture method (67. 7%). All the samples of control group including the organs of mice infected by Y. pseud otuberculosis and non-infected were negative by PCR method. The result showed that the PCR method is better than the traditional bacterial isolation and culture method ,for it is rapid,special and sensitive. So it is suitable for the surveillance of animal plague.
