LA辅助促排卵对金黄地鼠子宫内膜分泌与合成蛋白的影响

摘  要:对模仿人类体外受精(IVF)的动物模型金黄地鼠(hamster)的子宫内膜蛋白分泌及合成进行研究。结果表明:蛋白的分子量迁移在31~66KD,但蛋白电泳照片示在Leuprolide Acetate(LA)组其蛋白带密度明显低于孕马血清促性腺激素(PMSG)组;而新合成蛋白分析示在低分子和中分子量(31KD和45KD)LA组却明显高于PMSG组。且LA组分泌及合成蛋白均与无促排卵组(对照组)相似。该资料提示:使用LA(GnRHa类)促排卵使子宫内膜分泌趋向生理化是IVF高受孕率的机理之一,而子宫内膜蛋白分泌及合成受控于促排卵药物的种类。The present study was undertaken to evluate the endometrial secretion and synthesis of proteins in hamstrers who mimcked during in vitro fertilization (IVF) compared with the nondrug-treated hamsters. Uterine flushing protein patterns were analyzed by the technique of SDS-polyacrylamide gel electrophoresis (SDS-PAGE). Proteins synthesized were analyzed by liquid scintilation in radio-isotope counter. The results of protein resolution revealed that there three main protein patterns in which the molecular weights of the proteins migrated from 31KD to 66KD. Meanwhile, the intensities of protein bands in audoradiogran were higher in the hamsters treated with leuprolide acetate (LA) and pregnant mare's serum gondotropin (PMSG)than that in those treated with PMSG alone,but these was no significant difference between the intensi- ties of protein bands in groups LA + PMSA and saline. The results of determining newly synthesized [3H]-leucine labelled proteins showed that there was no difference between the couters in groups LA + PMSG and saline .but the amounts of low and middle molecular weights (31KD and 45KD)proteins in group PMSG were significantly less than that in groups LA + PMSG and saline. The data suggested that in the hamster,a physiologic endometrial protein secretory and synthetic changes with leuprolide acetate act as a possible cause of the higher pregnancy rate of IVF. The degree of endometrial secretion and synthesis varied considerably with the type of ovarian stimulation used.
